Advances in areas including genomics, automation and data science, as well as the diversification of therapeutic modalities, have led to a substantial expansion of the drug target landscape in the past 25 years. This Review maps and quantifies key elements of these changes, and discusses trends in the number and type of targets through which drugs mediate their therapeutic effects, which now include ~700 biomolecules.
